Community ecology across bacteria, archaea and microbial eukaryotes in the sediment and seawater of coastal Puerto Nuevo, Baja California

Microbial communities control numerous biogeochemical processes critical for ecosystem function and health. Most analyses of coastal microbial communities focus on the characterization of bacteria present in either sediment or seawater, with fewer studies characterizing both sediment and seawater together at a given site, and even fewer studies including information about non-bacterial microbial communities. As a result, knowledge about the ecological patterns of microbial biodiversity across domains and habitats in coastal communities is limited–despite the fact that archaea, bacteria, and microbial eukaryotes are present and known to interact in coastal habitats. To better understand microbial biodiversity patterns in coastal ecosystems, we characterized sediment and seawater microbial communities for three sites along the coastline of Puerto Nuevo, Baja California, Mexico using both 16S and 18S rRNA gene amplicon sequencing. We found that sediment hosted approximately 500-fold more operational taxonomic units (OTUs) for bacteria, archaea, and microbial eukaryotes than seawater (p < 0.001). Distinct phyla were found in sediment versus seawater samples. Of the top ten most abundant classes, Cytophagia (bacterial) and Chromadorea (eukaryal) were specific to the sediment environment, whereas Cyanobacteria and Bacteroidia (bacterial) and Chlorophyceae (eukaryal) were specific to the seawater environment. A total of 47 unique genera were observed to comprise the core taxa community across environment types and sites. No archaeal taxa were observed as part of either the abundant or core taxa. No significant differences were observed for sediment community composition across domains or between sites. For seawater, the bacterial and archaeal community composition was statistically different for the Major Outlet site (p < 0.05), the site closest to a residential area, and the eukaryal community composition was statistically different between all sites (p < 0.05). Our findings highlight the distinct patterns and spatial heterogeneity in microbial communities of a coastal region in Baja California, Mexico. En la Subcoordinación de Tratamiento de Aguas Residuales del IMTA se desarrolló un reactor biológico denominado BIOSTAR el cual ya está comercializado para el tratamiento descentralizado de aguas residuales de pequeñas poblaciones, fraccionamientos habitacionales, zonas residenciales, casas-habitación, hoteles, centros comerciales, edificios públicos, centros comerciales o recreativos. Se puede obtener agua con calidad adecuada para su desinfección con luz UV y posterior reutilización en servicios al público, cumpliendo con los límites máximos permisibles que para esto se establecen en la NOM-002-SEMARNAT/1997, así como con los límites para descarga a cuerpos receptores según la NOM-001-SEMARNAT/1996. El principal objetivo de tratamiento con el BIOSTAR es la remoción de la materia orgánica en el agua residual.

"En este artículo se estudia el doble o doppelgänger como tema central en La invención de Morel, de Adolfo Bioy Casares, a partir de los conceptos de doble objetivo y subjetivo (Jourde y Tortonese, 1996). El descubrimiento de la máquina de Morel incita un cambio de dobles en la novela: de uno objetivo a uno subjetivo. La identidad del narrador-fugitivo y su convivencia con las otras imágenes son elementos fundamentales para entender el papel del doble y su modificación en el relato. A pesar de haber sido mencionado por la crítica, ningún artículo había estudiado el doble como tema central de la novela, por lo cual esta investigación presenta una perspectiva innovadora. El uso de estos conceptos teóricos permite una aproximación a la complejidad narrativa del relato de Bioy".

Enhancement of antibiotics antimicrobial activity due to the silver nanoparticles impact on the cell membrane

The ability of microorganisms to generate resistance outcompetes with the generation of new and efficient antibiotics; therefore, it is critical to develop novel antibiotic agents and treatments to control bacterial infections. An alternative to this worldwide problem is the use of nanomaterials with antimicrobial properties. Silver nanoparticles (AgNPs) have been extensively studied due to their antimicrobial effect in different organisms. In this work, the synergistic antimicrobial effect of AgNPs and conventional antibiotics was assessed in Gram-positive and Gram-negative bacteria. AgNPs minimal inhibitory concentration was 10–12 μg mL-1 in all bacterial strains tested, regardless of their different susceptibility against antibiotics. Interestingly, a synergistic antimicrobial effect was observed when combining AgNPs and kanamycin according to the fractional inhibitory concentration index, FICI: &lt;0.5), an additive effect by combining AgNPs and chloramphenicol (FICI: 0.5 to 1), whereas no effect was found with AgNPs and β-lactam antibiotics combinations. Flow cytometry and TEM analysis showed that sublethal concentrations of AgNPs (6–7 μg mL-1) altered the bacterial membrane potential and caused ultrastructural damage, increasing the cell membrane permeability. No chemical interactions between AgNPs and antibiotics were detected. We propose an experimental supported mechanism of action by which combinatorial effect of antimicrobials drives synergy depending on their specific target, facilitated by membrane alterations generated by AgNPs. Our results provide a deeper understanding about the synergistic mechanism of AgNPs and antibiotics, aiming to combat antimicrobial infections efficiently, especially those by multi-drug resistant microorganisms, in order to mitigate the current crisis due to antibiotic resistance. © 2019 Vazquez-Muñoz et al.
