Chronic toxicity of shrimp feed added with silver nanoparticles (Argovit-4®) in Litopenaeus vannamei and immune response to white spot syndrome virus infection

"In recent years, the application of silver nanoparticles (AgNPs) as antibacterial compounds has been widely used in human and veterinary medicine. In this work, we investigated the effects of AgNPs (Argovit-4 R ) as feed additives (feed-AgNPs) on shrimp (Litopenaeus vannamei) using three different methods: 1) chronic toxicity after 28 days of feeding, 2) Effects against white spot syndrome virus (WSSV) challenged by oral route, and 3) transcriptional responses of immune-related genes (PAP, ProPO, CTL-3, Crustin, PEN3, and PEN4) following WSSV infection. The results showed that the feed-AgNPs did not interfere with the growth and survival of shrimp. Also, mild lesions in the hepatopancreas were recorded, proportional to the frequency of the feed-AgNP supply. Challenge test versus WSSV showed that feeding every 7 days with feed-AgNPs reduced mortality, reaching a survival rate of 53%, compared to the survival rates observed in groups fed every 4 days, daily and control groups of feed-AgNPs for the 30%, 10%, and 7% groups, respectively. Feed-AgNPs negatively regulated the expression of PAP, ProPO, and Crustin genes after 28 days of treatment and altered the transcriptional responses of PAP, ProPO, CTL-3, and Crustin after WSSV exposure. The results showed that weekly feeding-AgNPs could partially prevent WSSV infection in shrimp culture. However, whether or not transcriptional responses against pathogens are advantageous remains to be elucidated."

Hábitos alimenticios y aportación nutricional de la dieta de Paralichthys californicus, en la Bahía de Todos Santos y estero de Punta Banda, Ensenada, B.C

Se analizaron 289 estómagos para describir la dieta de Paralichthys californicus en la Bahía de Todos Santos y el Estero de Punta Banda (junio de 1992 a marzo de 1993), realizando una separación por área y estación (invierno y verano). Se determinó realizar la descripción de la dieta a nivel de grupos presa para categorizar las presas ingeridas, y se emplearon los índices de porciento numérico, de peso, de frecuencia de ocurrencia, y el Índice de Importancia Relativa (IIR). Los organismos de la bahía presentaron mayor talla promedio (154.2 ±D.E.37.2mm) que los colectados en el estero (107.9 ±36.85mm), la cual fue diferente (K-W, p<0.001) tanto entre las dos áreas como entre invierno y verano de cada zona. En ambos lugares los organismos de menor talla se encontraron en invierno. Se identificaron cuatro presas a nivel familia, ocho a nivel género, y 15 a nivel especie. Los principales componentes de la dieta en la bahía durante invierno, fueron mysidos con 88.4% del I.I.R. (Metainysidopsis elongata, 62. l %), carideos (10%) y peces; en el verano el grupo más importante nuevamente fue mysidos (98.7%), y el resto gamáridos, carideos y peces. De las presas presentes en el estero, los peces fueron los más importantes (95.6%) en el invierno destacando góbidos (60.63%) y principalmente Ilypnus gilberti (30.5%); en el verano, varias especies de góbidos 31.68% y otras especies de peces (35.2%). Se encontraron diferencias (p<0.01) en el número de presas tanto entre las dos áreas como entre el invierno y el verano de ambos lugares. En el estero, se detectaron diferencias en el porcentaje de proteínas de los contenidos estomacales en los dos grupos de tallas y estaciones del año; un resultado similar fue para lípidos. Una estimación de las calorías con base en el contenido de lípidos y proteínas de las principales presas, indicó que el aporte calórico al depredador está en función del número y tipo de presa ingerida y no a la selección de las presas por el lenguado, esto puede reflejar la disponibilidad del alimento. Los valores de ración diaria fueron mayores para la bahía (0.22-0.24 Kcal/g) que para el estero (0.10-0.12 Kcal/g), y no se encontró un efecto del cambio de temperatura entre verano e invierno (Q10 <1.4) para ambos lugares.

A total of 289 stomachs were analyzed to describe the diet of Paralichthys californicus juveniles in Bahía de Todos Santos and Estero de Punta Banda, B.C., México (June 1992 to March 1993); after the collection, the stomachs were grouped in winter and summer seasons of each area. Diet was described to higher-1evel taxonomic categories based on the percent in number, wet weight, and frequency of occurrence and the prey importance by the Index of Relative Importance (IIR). Largest organisms (154.2± D.E.37.2mm SL) were found in the hay than those from the estuary (107.9±.36.85mm), with a significative difference between areas and between winter and summer of each site (K-W, <0.001) In both areas the minor size organisms were collected during winter. The lowest taxon identified as preys in both areas were four at family leve1, and eight to genus and 15 to species level. Mysids (IRI = 88.4%) with Metamysidopsis elongata (62.1 %), carideans (10.2%), and fishes were the most important food items in the bay during winter; in summer, the mysids (IRI = 98.4%, M. elongata, IRI = 35.3%), carideans (0.9%) and the fishes, again. ln the estuary (winter), the rnost important were the fishes (IRI = 95.6%) with members of the gobies (60.63%) mainly llypnus gilberti (30.5%); a small percent was provided by invertebrates (0.08%); in summer, the fishes (66.9%), gamarids (17%) and the mysids ( 13. 8%) domínate the diet. Significant differences was found in prey numbers between areas and between seasons of each site. In the estuary, differences in the protein content from stomachs of two groups size were found; these differences were detected between seasons, and similar results was detected in lipids. The calories values (based in lipid and protein content) from the main prey, indicate that the caloric contribution is a function of kind and prey numbers, more than the selection by the halibul, this may indicate the food disponibility. The daily ration in bay (0.22-0.24 Kcal/g) was greater than in the estuary (0.10-0.12), and no effect of the temperature change in summer and winter (Q10 <1.4) for both areas were found.

El aprendizaje por refuerzo es un paradigma del aprendizaje de máquina con un amplio desarrollo y una creciente demanda en aplicaciones que involucran toma de decisiones y control. Es un paradigma que permite el diseño de controladores que no dependen directamente del modelo que describe la dinámica del sistema. Esto es importante ya que en aplicaciones reales es frecuente que no se disponga de dichos modelos de manera precisa. Esta tesis tiene como objetivo implementar un controlador óptimo en tiempo discreto libre de modelo. La metodología elegida se basa en algoritmos de aprendizaje por refuerzo, enfocados en sistemas con espacios de estado y acción continuos a través de modelos discretos. Se utiliza el concepto de función de valor (Q-función y función V ) y la ecuación de Bellman para resolver el problema del regulador cuadrático lineal para un sistema mecánico masa-resorte-amortiguador, en casos donde se tiene conocimiento parcial y desconocimiento total del modelo. Para ambos casos las funciones de valor son definidas explícitamente por la estructura de un aproximador paramétrico, donde el vector de pesos del aproximador es sintonizado a través de un proceso iterativo de estimación de parámetros. Cuando se tiene conocimiento parcial de la dinámica se usa el método de aprendizaje por diferencias temporales en un entrenamiento episódico, que utiliza el esquema de mínimos cuadrados con mínimos cuadrados recursivos en la sintonización del crítico y descenso del gradiente en la sintonización del actor, el mejor resultado para este esquema es usando el algoritmo de iteración de valor para la solución de la ecuación de Bellman, con un resultado significativo en términos de precisión en comparación a los valores óptimos (función DLQR). Cuando se tiene desconocimiento de la dinámica se usa el algoritmo Q-learning en entrenamiento continuo, con el esquema de mínimos cuadrados con mínimos cuadrados recursivos y el esquema de mínimos cuadrados con descenso del gradiente. Ambos esquemas usan el algoritmo de iteración de política para la solución de la ecuación de Bellman, y se obtienen resultados de aproximadamente 0.001 en la medición del error cuadrático medio. Reinforcement learning is a machine learning paradigm with extensive development and growing demand in decision-making and control applications. This technique allows the design of controllers that do not directly depend on the model describing the system dynamics. It is useful in real-world applications, where accurate models are often unavailable. The objective of this work is to implement a modelfree discrete-time optimal controller. Through discrete models, we implemented reinforcement learning algorithms focused on systems with continuous state and action spaces. The concepts of value-function, Q-function, V -function, and the Bellman equation are employed to solve the linear quadratic regulator problem for a mass-spring-damper system in a partially known and utterly unknown model. For both cases, the value functions are explicitly defined by a parametric approximator’s structure, where the weight vector is tuned through an iterative parameter estimation process. When partial knowledge of the dynamics is available, the temporal difference learning method is used under episodic training, utilizing the least squares with a recursive least squares scheme for tuning the critic and gradient descent for the actor´s tuning. The best result for this scheme is achieved using the value iteration algorithm for solving the Bellman equation, yielding significant improvements in approximating the optimal values (DLQR function). When the dynamics are entirely unknown, the Q-learning algorithm is employed in continuous training, employing the least squares with recursive least squares and the gradient descent schemes. Both schemes use the policy iteration algorithm to solve the Bellman equation, and the system’s response using the obtained values was compared to the one using the theoretical optimal values, yielding approximately zero mean squared error between them. An adaptability test is conducted considering variations that may occur in plant parameters, with the least squares with recursive least squares scheme yielding the best results, significantly reducing the number of iterations required for convergence to optimal values.

Genetic analysis of Vibrio parahaemolyticus O3:K6 strains that have been isolated in Mexico since 1998

Vibrio parahaemolyticus is an important human pathogen that has been isolated worldwide from clinical cases, most of which have been associated with seafood consumption. Environmental and clinical toxigenic strains of V. parahaemolyticus that were isolated in Mexico from 1998 to 2012, including those from the only outbreak that has been reported in this country, were characterized genetically to assess the presence of the O3:K6 pandemic clone, and their genetic relationship to strains that are related to the pandemic clonal complex (CC3). Pathogenic tdh+ and tdh+/trh+ strains were analyzed by pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST). Also, the entire genome of a Mexican O3:K6 strain was sequenced. Most of the strains were tdh/ORF8-positive and corresponded to the O3:K6 serotype. By PFGE and MLST, there was very close genetic relationship between ORF8/O3:K6 strains, and very high genetic diversities from non-pandemic strains. The genetic relationship is very close among O3:K6 strains that were isolated in Mexico and sequences that were available for strains in the CC3, based on the PubMLST database. The whole-genome sequence of CICESE-170 strain had high similarity with that of the reference RIMD 2210633 strain, and harbored 7 pathogenicity islands, including the 4 that denote O3:K6 pandemic strains. These results indicate that pandemic strains that have been isolated in Mexico show very close genetic relationship among them and with those isolated worldwide. © 2017 Guerrero et al. Phylogenetic relationships of Pseudo-nitzschia subpacifica (Bacillariophyceae) from the Mexican Pacific, and its production of domoic acid in culture

Pseudo-nitzschia is a cosmopolitan genus, some species of which can produce domoic acid (DA), a neurotoxin responsible for the Amnesic Shellfish Poisoning (ASP). In this study, we identified P. subpacifica for the first time in Todos Santos Bay and Manzanillo Bay, in the Mexican Pacific using SEM and molecular methods. Isolates from Todos Santos Bay were cultivated under conditions of phosphate sufficiency and deficiency at 16°C and 22°C to evaluate the production of DA. This toxin was detected in the particulate (DAp) and dissolved (DAd) fractions of the cultures during the exponential and stationary phases of growth of the cultures. The highest DA concentration was detected during the exponential phase grown in cells maintained in P-deficient medium at 16°C (1.14 ± 0.08 ng mL-1 DAd and 4.71 ± 1.11 × 10−5 ng cell-1 of DAp). In P-sufficient cultures DA was higher in cells maintained at 16°C (0.25 ± 0.05 ng mL-1 DAd and 9.41 ± 1.23 × 10−7 ng cell-1 of DAp) than in cells cultured at 22°C. Therefore, we confirm that P. subpacifica can produce DA, especially under P-limited conditions that could be associated with extraordinary oceanographic events such as the 2013–2016 "Blob" in the northeastern Pacific Ocean. This event altered local oceanographic conditions and possibly generated the presence of potential harmful species in areas with economic importance on the Mexican Pacific coast. © 2020 Quijano-Scheggia et al.
